A method for preparing an antigen comprising a peptide-protein complex which comprises reacting a peptide represented by the general formula (I) <IMAGE> (I) wherein m is 0 or 1, as a hapten, with a protein as a carrier in the presence of a dialdehyde represented by the general formula (II) OHC-(CH2)n-CHO (II) wherein n is an integer of 1 to 5 and a method for preparing an antibody specific to pancreatic glucagon comprising administering the above described antigen to a mammal and recovering the antibody produced.
