PURPOSE:To detect accurately and with good reproductivity the endotoxine in a specimen by using a limulus amebocyte lysate component by means of using a specimen that is a living body specimen treated by acid under the condition that limulus solidification type enzyme substrate cutting activity has become not to be detected. CONSTITUTION:A body liquid specimen such as blood, abdominal dropsy, urine, etc. is treated at a pH value under 3 with the existence of an acid such as mono-, di-, and tri-chloracetic acids that has pKa value under 3 at 25 deg.C, or a strong acid such as chloric acid, perchloric acid, hydrochloric acid, sulfuric acid, etc. in order to adjust the specimen so that the limulus solidification type enzyme substrate cutting activity is not detected. To this specimen a limulus amebocyte lysate component is added to measure the solidification reaction. With this arrangement is becomes possible to detect exactly endotoxine in a living body specimen by dissolving or removing obstructing substances for endotoxine that are proteins, cell granules, etc. with adsorptivity to endotoxine without giving unfavorable effects to the endotoxine. |
