A reagent for use in the TPHA method for the diagnosis of syphilis which reliably enables syphilis to be detected in the primary stage comprises antigen obtained from fractions of a culture of Treponema pallidum Nichols having a specific gravity of at least 1.01 sensitising of carrier particles, which antigen is to undergo hemagglutination in the presence of antibody.
