PURPOSE:cDNA producing tropoelastin useful for inhibitor of blood platelet aggregation preventing arteriosclerosis, obtained by coding tropoelastin. CONSTITUTION:A cell producing a large amount of elastin, derived from chicken, is ground to give a RNA fraction, from which polyA<+>RNA (A) is isolated. Then the component A is treated with a restriction enzyme to give cDNA-containing plasmid (B). Then the component B is transformed with Escherichia coli to give plasmid recipient bacterium (C). On the other hand, the component A is linked to polyA<+>RNA obtained from an elastin nonforming cell and labeled with <32>P, etc., to give <32>P labeled polyA<+>RNA probe (D). Then the components D and C are hybridized, a colony showing complementation to a probe derived from the component A and plasmid DNA is extracted from the clone of the prepared colony. Then the DNA is treated with a restriction enzyme to give cDNA of tropoelastin showing base sequence such as peptide shown by the formula and crosslinking forming peripheral sequence, etc. |
