NEW MATERIAL:A surface antigen of hepatitis B virus, expressed by the formula (HBs antigen). USE:A diagnosticum or a vaccine for hepatitis B virus infection. PREPARATION:For example, a serum of a patient infected by hepatitis B virus (HBV) is centrifugated at 10,000rpm to separate a supernatant liquid. This supernatant liquid is further centrifugated at 28,000rpm for 4hr and HBV virus particles are obtained from the sediment. The virus particles are treated with a protease, etc., and further treated with a water saturated phenol to obtain DNAs. Then the DNAs are treated with restriction endonucleases, subjected to cloning and then combined with a replicable expression vector to form a recombinant DNA. The recombinant DNA is then introduced into a microorganism to transform, and this transformant is cultured to obtain the compound expressed by the formula from the culture fluid. |
