PURPOSE:To obtain an antigen suitable for gaining an antibody used in the immunological analysis of oestriol with a high accuracy by combining bovine serum albumin to a position C6 of B ring of a 6-oxoestriol as starting material, not to a glucuronic acid moiety of D ring. CONSTITUTION:After OH at C3 of A ring in 6-oxoestriol of formula III is changed to C6H5CH2O (benzyl eter) group, H of OH at C16 of D ring is replaced by a glucuronyl group having acetyl groups in positions C2, C3 and C4 of formula IV. Then, after the reduction of C6H5CH2O to H, NOCH2COOH is introduced into position C6 of B ring. Then, after the reaction with bovine serum albumin (BSA), a hydrolysis is conducted to convert the acetyl group into OH. Thus, an antigen is obtained for assaying oestriol-16alpha-dlucuronide of formulaI(where, G represents the glucuronyl group of formula II). This antigen is injected into a rabbit to obtain an antiserum. The use of the antiserum enables a highly sensitive assaying of oestriol in plasma and urine without the hydrolysis of the plasma and the urine. |
