PURPOSE:A novel plasmid that is used as a vector plasmid to introduce an additional exogenote into cells of Escherichia coli into which an exogenote has been introduced by use of a colicin E1 vector. CONSTITUTION:A plasmid that can have coexistence in the same host cells with a colicin E1 plasmid such as pBR322 or a recombinant DNA prepared by use of the same plasmid. For example, pGA plasmid is segregated with restriction enzyme BamHI to obtain a reaction mixture containing a couple of DNA fractions with 2.8X10<3> base pair and 5.1X10<3> base pairss respectively and the mixture is treated with T4 ligase to effect the binding reaction to prepare the objective pGB1 plasmid of a cyclic DNA with 2.8X10<3> base pair. |
