PURPOSE:To determine phenols quantitatively with high sensitivity, by allowing 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) to react with phenols and measuring colored fluorescence arising by a reacted solution. CONSTITUTION:A detection reagent is made by dissolving 0.02-2% DDQ shown by a formula in a mixed solvent e.g. toluene/acetic acid (6:4 volume ratio). Phenols extracted from a sample solution by a suitable method are added to said reagent solution. Colored fluorescence arising by reaction in accordance with phenols, is measured by a fluorescence spectrophotometer or ultraviolet spectrophotometer and the phenols are determined quantitatively in comparison with a preliminarily determined calibration curve with high accuracy. For example, n-butanol and salt are added to urine of a pregnant woman under a condition which is made acidic with hydrochloric acid and are mixed thoroughly. Hereafter, said mixture is centrifuged and the frozen and separated butanol layer is developed into a thin layer plate and then, said plate is dried by air. Further, the reagent solution is atomized and estroil in the urine of the pregnant woman is measured by measuring its coloration spectrally. |
