NEW MATERIAL:The 27-desamidosecretin of formula (Val-OH represents that the C-terminal of the polypeptide composed of valine is carboxylic acid). USE:It exhibits the action to promote the secretion of the pancreatic juice by intravenous injection, and is useful as a substance having a bioactivity similar to secretin. It can be used also as secretin by amidating the carboxyl group at the C-terminal. PROCESS:The structural gene of desamidosecretin corresponding to the polypeptide having secretin structure wherein the C-terminal amino acid is valine, is synthesized chemically, and integrated into the plasmid which can be proliferated in the projected host cell (e.g. colon bacilli belonging to Escherichia genus) to produce chimera plasmid proliferable in the cell. The host cell is transformed by the plasmid, the resultant transformation is cultured and the produced desamidosecretin is recovered. |
