PURPOSE:To obtain a result of analysis having good reproducibility in good accuracy, by classifying a sample into each group of bile acid of isolation type, glycine combination type and taurine combination type and analysing each group by using a moving phase of the same composition under the same condition. CONSTITUTION:After desalting and protein removing treatment of a sample is carried out, obtained eluate is treated by hydrophobic gel having an ion exchange radical and is classified into three fractions of bile acid containing isolation type, glycine combination type and taurine combination type. Further, these three fractions are treated by a high speed chromatography using the same moving phase under the same condition and the obtained eluate is mixed with a reaction reagent. Next, a enzyme catalytic reaction is carried out by passing through a fixation enzyme column and the obtained reacted solution is treated by fluorescence analysis and then, each bile acid is analyzed. Hereby, the result of analysis having good reproducibility is obtained without fluctuation of a base line. |
