PURPOSE:To slow down unknow substances in a moving phase and raise detection sensitivity by providing between the liquid conveying section and the specimen charging section a play column that has an internal volume that is more than 60% of the separation column and is packed with strong acidic cation exchange resin. CONSTITUTION:Cleaning liquid 26 is made to flow from the liquid conveying section 21 to clean, and after a first moving phase 23 is made to flow, the specimen is poured from a specimen supply section 13. Further, a first, second and third moving phases 23-25 with different pH's are made to flow to promote aminoacid separation in the separation column 14. Into the column 14 an oxidizing agent 21 of low concentration is continuously poured from an oxidizing reagent supply section 15 to effect reaction in a first reaction section 16 and also fluorescent reagent 22 that contains a large volume of non-ion surface active agent is poured from a fluorescent reagent supply section 17 to effect reaction in a second reaction 18. The intensity of fluorescence is measured and aminoacid is analysed. Aminoacids can be analysed with high accuracy by delaying the movement of ammonia and other unknown substances in the moving phases more than the most delayed aminoacids by the play column 20. |
