PURPOSE:A strain producing a novel antibiotic in a actinomadura is cultured and the titled substance used as an active ingredient in medicines, because of its activity of inhibiting the growth of gram-positive bacteria, is collected from the culture mixture. CONSTITUTION:A strain FA-1180 (FERM-5155) in actinomadura is cultured in a usual medium for actinomycetes containing carbon source, nitrogen source, growth accelerator, inorganic salts and others at about 20-40 deg.C and 4-9pH for 7-20 days. The resultant culture mixture is filtered and the filtrate is extracted. The objective substance is separated, collected and purified by utilizing the difference of the antibiotic in adsorptivity or affinity to adsorbents and in solubility in solvents. The resultant antibiotic substance is amphoteric, dark-red needle, melts at 152-154 deg.C and shows Rf=0.45 in the silica-gel thin-layer chromatography developed with chroloform, toluene and methanol.
|
