PREPARATION OF RECOMBINED DNA

PURPOSE:A recombined DNA is prepared by use of a DNA of a bacteriophage in a host of actinomycetes that has only one position cleavable with a restricted enzyme. CONSTITUTION:A restricted enzyme, Pvu II is made to act on the DNA of temperature bacteriophage in a host of actinomycetes to cleave it and the product is mixed with an extrinsic DNA having flash end. Then, a DNA-ligase is made to act thereon to give the objective recombined DNA. The DNA of temperate bacteriophage R4 is cleaved at only one position by restricted enzyme Pvu II and the position is different from that bearing genetic information.
