PURPOSE:To prepare tryptophanase in high yield, by cultivating a bacterium capable of producing tryptophanase in a medium containing L-glutamic acid or its salt. CONSTITUTION:A well-known bacterium belonging to the genus Proteus, Escherichia, etc., capable of producing tryptophanase is inoculated into a nutrient medium containing 0.5-2wt% L-glutamic acid or its salt such as sodium L- glutamate and cultivated by shaking culture preferably at a pH of 4-8 at 20- 40 deg.C, tryptophanase is collected from the culture mold and treated with indole and L-serine or indole and ammonium puruvate, to give L-tryptophan. |
