<p>PURPOSE:To conduct separation of Serum that is required for clinical test in a short time and with high recovery without hemolysis of red blood corpuscles, by adding a given fraction of babu poison or a purified product thereof in a small quantity to human blood. CONSTITUTION:Dried crude babu poison is dissolved in water and lipid is extracted with an organic solvent. An aqueous layer is dialyzed and the dialyzate is allowed to be adsorbed on a Sephadex column and eluted with a buffer solution to obtain a fraction having a strong blood coagulation activity. This fraction is freeze-dried ro obtain an enzyme having a thrombin-like activity. A required quantity of the enzyme is dissolved in physiological saline solution and used as a sterilize coagulating enzyme solution. 1ml of the enzyme solution thus obtained contains 100mug of sterlize coagulating enzyme, 1mg of gelatin hydrolyzate, 8.76mg of sodium chloride, and 50mg of glycine. It can be stored as such or by freeze drying as a blood coagulation promoting agent.</p> |
