<p>PURPOSE:To prepare the titled compound useful as a synthetic raw material of pesticides, pharmaceuticals, and alkaloids, easily, in high yield, by reacting 4- amino-1-methoxy-carbonyl-indole with NaNO2 in the presence of an inorganic acid. CONSTITUTION:The objective 4-hydroxy-1-methoxycarbonyl-indole can be prepared in high yield, by reacting 4-amino-1-methoxycarbonyl-indole of formulaI with NaNO2 at 0-30 deg.C in the presence of an inorganic acid (e.g. hydrochloric acid, sulfuric acid, etc.), and thermally decomposing the resultant diazonium salt at 70-100 deg.C for >=30min. The starting compound of formulaIis obtained by reacting 4-nitro-1-hydroxy-indole of formula III with the halogen-substituted acetic acid ester of formula IV (X is halogen; R is lower alkyl) to obtain 4-nitroindole of formula V in high yield, reacting the product with methyl chloroformate, and reducing the resultant compound of formula VI.</p> |
