PURPOSE:To detect cyclic amino acids in particular with high sensitivity by oxidizing the amino acid eluate separated with a liquid chromatograph with NaClO, etc. in the 1st reaction part, then causing the same to react with o- phthalaldehyde in the 2nd reaction part and subjecting the reacting liquid to fluorescence analysis. CONSTITUTION:In an amino acid analysis device 1, sample is put into a separating column 6 from a sample introducing device 5, and a mobile phase 21 (the one contg. sodium citrate, n-caprylic acid and ethanol and controlled to 3.25pH with perchloric acid) is fed into the column with a pump 4. The liquid contg. 10<-12>-10<-8>mol amino acid eluted from the column 6 is caused to react with the reagent 9 prepd. by adding an oxidizing agent such as NaClO into a buffer soln. of 10.2pH consisting of Na2CO3-HBO3-K2SO4 to 0.1% concn. in a reaction tube 12. Next, the reagent 15 prepd. by dissolving o-phthalaldehyde to 0.2-0.5% concns. in the same buffer soln. as mentioned above is fed at 0.2-0.5pts.vol. based on 1pt.vol. the reacting liquid obtained from the reaction coil 12. The fluorescence emitted by this is measured with a detector 19. Thereby, the cyclic amino acids such as proline are made measurable with high sensitivity. |
