PURPOSE:To obtain a reagent which enables precise and handy analysis, by introducing phospholipid, glycolipid or fatty acid with an antigen or an antibody immobilized thereon and a label substance simultaneously after the process of forming films of phospholipid and glycolipid. CONSTITUTION:First, a moderate amount of lipid and of cholesterol as required is put into a flask, dissolved and mixed into a solvent added thereto and then, after the removal of the solvent by suction, when the work is dried, a lipid thin film is formed on the wall surface of the flask. On the other hand, an antigen or antibody is brought to a chemical reaction with phospholipid, glycolipid or a derivative of fatty acid to prepare an antigen or antibody immobilized on the phospholipid, glycolipid or the derivative of fatty acid. Then, a solution containing a label substance and the antigen or antibody immobilized on the phospholipid, glycolipid or the derivative of fatty acid are added into the flask on the wall surface of which is formed a lipid thin film and sealed with a stop, the flask is shaken to prepare a suspension of liposome. The liposome herein used may be in either form of a large single-layer or multi-layer film.
