Spiro-hydantoins I (X = O, S, SO, SO2; R,R1 = H, F, Cl, Br, lower alkyl, alkoxy) were prepd. Thus, a mixt. of 2.5 g 6- methoxy -1-indanone, 1.5 g KCN and 6.7 g (NH4)2CO3, which was dissolved in 20 ml EtOH, was heated to 110≦̸C for 20 hr, cooled to room temp. and adjusted to pH 2.0 with 6N HCl, the resulted precipitates were recrystallized with EtOH to give 0.49 g 6'-methoxy -spiro-[imidazolydin-4,1'-indan -2,5-dione (m.p. 192-4≦̸C). I inhibited aldose reductase activity, decrease sorbitol accumulation, and inhibited cataract formation in diabetics.
