<p>NEW MATERIAL:A compound of formula I[R1 and R3 are H or protecting groups; R2 is oxo group (=0), OH or protected OH group]. EXAMPLE:22-Cyano-3B, 25-dihydroxy-24-oxocholest-5-ene. USE:An intermediate of a compound for a haptene usable in the enzymic immunoassay. PROCESS:A 24-oxochloesta-5,22-diene of formula II is reacted with a cyanide, preferably potassium cyanide, in an equivalent amount of preferably 1.5-3 times that of the diene in a mixed solvent of water with DMF at 5-50 deg.C for 0.5-24hr to give a compound of formula III in which groups R2 in formula I is oxo group. The starting raw material compound of formula II is obtained as follows: A 3-methyl-3-(tetrahydropyran-2-yloxy)butan-2-one is reacted with a 3beta-acetoxy- 23, 24-bisnorchol-5-enal, and the reaction product is then treated with an acid to give the compound of formula II in which R1 is acetyl and R3 is H.</p> |
