A plasminogen activator is isolated by (i) treating stromata of erythrocytes, preferably human, with an aqueous acidic solution, e.g. aqueous HC1 or H 2 SO 4 ; (ii) extracting the activator from the aqueous solution into an organic phase; and (iii) isolating the activator from the organic phase. After step (i), and before step (ii), insoluble solids may be removed from the solution, and/or the solution may be dialysed to remove dialysable impurities. The activator may be extracted from the aqueous solution into an organic phase comprising or consisting of a phenol and/or a chlorophenol and subsequently extracted from the organic phase into aqueous solution by the addition of water and an excess of a non-polar organic solvent, e.g. isopropyl ether-chloroform. The activator may be further purified by dialysis, adsorption, chromatography, isoelectrical precipitation or gel filtration and the activator may be isolated by lyophilization or precipitation. The plasminogen activator may be used in the treatment and prophylaxis of thromboses and embolisms in the form of an injectable solution.
