NEW MATERIAL:An insulin analog in which the amino acid at the 30-position of the B chain is substituted by a >=5C amino acid or a derivative thereof. EXAMPLE:[B30-Leucine]-hog insulin. USE:A remedy for diabetes, particularly useful as a remedy for an insulin-resistant diabetic patient (a patient to whom the remedy is not suitable due to the antibody held against the bovine or hog insulin thereof). PROCESS:Des-B30-insulin prepared from various animal insulins is condensed with a >=5C D- or L-amino acid or a derivative thereof to give the aimed insulin analog. The condensing reaction is carried out in the presence of a protease, e.g. trypsin, having the ability to cleave the carboxyl group side of the basic amino acid residue. |
