PURPOSE:To obtain a novel L-arginine oxydase from the cultured product of bacteria belonging to Arthrobacter genus. CONSTITUTION:Arthrobacter sp.B-790 (FERM-P No.5663) is cultured at 25- 30 deg.C for 24-72hr in a conventional aerated and agitated deep-tank fermentor, and the bacterial cells are separated from the medium and disintegrated to obtain crude enzyme solution. The objective L-arginine oxydase is prepared from the solution by fractional precipitation, chromatographic purification, and freeze-drying. The enzyme has a molecular weight of 140,000, Km value of 25X10<-3>M, and an isoelectric point of about pH. 4.81, catalyzes the reaction of formula (I), and has the optimum pH of about 6.5. The Arthrobacter sp. B-780 strain is similar to Arthrobacter citreus, but different in the bacteriological characteristics. |
