<p>PURPOSE:To obtain undecapeptide suitable for preparing a specific antibody useful for measuring the concentration of pancreas glucagon in blood, by condensing a hydrazide compound with heptapeptide by azide method. CONSTITUTION:Benzyloxycarbonyl-alanyl-glutaminyl-asparaginyl-phenylala nine hydrazide is reacted with isoamyl nitrite in the presence of hydrochloric acid to give an azide, which is reacted with a heptapeptide obtained by reacting tertiary butyloxycarbonyl-valyl-glutaminyl-tryptophyl-leucine hydrazide with methionyl- asparaginyl-threonine by azide method, to give an undecapeptide having a protecting group. The undecapeptide is reacted with sodium in liquid ammonia and the protecting group is removed to give the desired undecapeptide. This undecapiptide is condensed with bovine serum albumin, etc. to give an antigen, from which an antibody having high specificity can be obtained.</p> |
