PURPOSE:To detect occult blood in a sample after a long-time preservation, by detecting hemoglobin and transferrin simultaneously by immunological measurement after a glycosidase type bacteriolysis enzyme acts on a sample excrement. CONSTITUTION:An excrement sampling spoon is immersed into a buffer containing an anti-human hemoglobin antibody and an anti-human transferrin antibody and dried after left alone for one night. The spoon sticks into excrements to sample a fixed amount thereof. On the other hand, a bacteriolysis enzyme such as lysozyme and NaN3 are added to agar solution to gelatinize. The sampling spoon is inserted into the agar gel and left alone for a fixed time. Then, the spoon is taken out to wash and immersed into an alkali phosphatase label antibody solution. Then, the spoon is washed, immersed into a phenyl phosphate substrate liquid to incubate and after the addition of a coloring liquid, reaction is stopped to measure the degree of coloration. |
