PURPOSE:To prepare the titled substance useful as a raw material of pharmaceuticals, economically, by culturing microorganisms belonging to Corynebacterium genus in a medium containing a cholic acid salt. CONSTITUTION:Microorganisms belonging to Corynebacterium genus, e.g. Corynebacterium CA-53 (FERM-P No.5532), etc. are cultured in a medium containing a cholic acid salt as a substrate. The medium contains a carbon source, a nitrogen source, inorganic salts, etc. assimilable with the microorganisms. The cholic acid salt is, e.g. an alkali metal salt. The culturing is carried out under aerobic conditions, and after the completion of the cultivation, the objective 3-substituted-7alpha- hydroxy-12-keto-5beta-cholanic acid and/or its salt are obtained by the removal of the cells, addition of hydrochloric acid, addition of ethyl acetate, etc. |
