A process for preparing alkoxychlorodialkylaminophosphine comprises (a) reacting trichlorophosphine with alcohol in an aprotic solvent at -70-0 deg.C to obtain dichloromethoxyphosphine, (b) evaporizing HCl and the solvent at 30-50 deg.C, and (c) adding dialkylamine to the reaction soln. at -70-0 deg.C. Pref. the alcohol is methanol, ethanol, diisopropanol, butanol or beta-cyanoethanol, and the dialkylamine is morpholine, diethylamine, dimethylamine or diisopropylamine. Phosphoramidite is useful in the chemical synthesis of DNA.
