is obtd. by culturing Streptomyces hygroscopicus 9735-1 in a suitable nutrient medium under aerobic conditions. The pH at the beginning of cultivation is pref. 6.5-7.5. and the cultivation temp. is pref. 27-31 degrees C. The amt. of DE-3936 accumulated reaches the max. in 65-110 hrs. cultivation. Recovery of the DE-3936 can be effected conventionally. Since the DE-3936 (in the form of free acid or its salt) is soluble in various organic solvents, the culture is treated with a suitable solvent, and the extract is then subjected to chromatography or recystallisation. Since the DE-3936 is contained in both the cultivated cells and culture filtrate, the culture is first sepd. into the cells and the filtrate. The cells are treated with methanol or acetone, and the filtrate is treated with benzene, chloroform, dichloroethane or ethyl acetate. DE-3936 in the form of sodium salt is obtd. as colourless prisms, and the structure is represented by the following formula: DE-3936 is effective for malaria, toxoplasma or coccidium infection and partic. effective against protozoa which are resistant to the currently used sulfa type drugs, quinoline type drugs or antithiamine. |
