PURPOSE:To measure concentration of a substrate in a liquid to be measured, by a method wherein hydrogen peroxide, which is produced resulting from the reaction of the substrate due to the action of ferment, is reacted with iodic ion in an electrolytic liquid to prepare iodine, and the iodine is detected polargraphically. CONSTITUTION:A ferment fixed film 6, in which ferment, such as glucoseoxidase, is fixed, is positioned at a forward end of a body 3, and a cathode 5, consisting of a platinum electrode, makes contact with the inside of the fixed film 6 through the medium of a cellophane 7. An anode 2 consists of a silver-iodine silver electrode, and the electrode is filled with a buffer solution, whose pH is 4-7, and an electrolytic liquid 4 loaded with iodide calium and metal oxide. A direct current electrolytic voltage from a power souce 1 is applied between the anode 2 and the cathode 5. Hydrogen peroxide, generated resulting from the reaction of the substrate, reacts with iodide calium through the aid of the metallic oxide, acting as a catalyst, in the electrolytic current lows between the anode 2 and the cathode 5. Iodine predischarge as compared with oxygen, and thereby it is not interfered with oxygen and other components. |
