PURPOSE:To satisfy a high separation capacity, a high-speed processing, and a high durability simultaneously, by a method wherein factors, of which a filling column consists, are stuided synthetically, and both gel property and a filling property are displayed to the maximum. CONSTITUTION:In an organic solvent gel permeation chromatography column in which a total porous filler, consisting of mainly organic synthetic polymer, is used, the filler is prepared such that a containing solvent amount of gel is 1.0-3.0ml/ g, and a gel particle internal capacity/ a gap capacity between gel particles is 1.3- 1.7. The particle diameter of the gel is average 5-30mum, preferably 5-15mum, and a theoretical plate equivalent height/mean gel diameter is 5.0 or less, preferably 3.0 or less. Furthermore, the particle distribution of the gel is preferably 1.5 or less in weight mean diameter/number mean diameter. A preferable column is formed in a cylinder which measures 4-50mm. in an inner diameter and 1,000mm. or less in the length. Both ends of a gel bed are fastened by means of end fittings providing with a screen through which liquid is caused to pass but the gel is caused not to pass. |
