PURPOSE:The strain of Bacillus stearothermophilus UK788 is cultured batchwise to the late period of the logarithimic growth step to prepare the titled dehydrogenase with high thermal stability and high activity kept for a long period of time. CONSTITUTION:The strain of Bacillus stearothermophilus UK788 is aerobically cultivated in a usual medium that is used to cultivate a bacterium. When the cultivation is conducted batchwise, it is preferably continued to the late period of the logarithmic growth step or in the continuous cultivation, the strain is cultured so that the dilution rate is controlled near the maximum of specific growth rate of the UK788 strain. The objective heat-resistant glucose-6-phosphate dehydrogenase is isolated and purified from the culture mixture by collecting the cells filtration or similar operation and effecting usual fractionation. |
