PURPOSE:A microorganism belonging to Bacillus is cultured and the culture mixture is purified with a combination of specific column chromatographies to produce high- purity heat-resistant glucokinase in high efficiency. CONSTITUTION:A microorganism belonging to Bacillus, e.g., Bacillus stearothermophilus is inoculated in an enriched culture medium and incuvated under aerobic conditions. The culture mixture is subjected to usual extraction fractionation to separate crude fractionated extracts. The crude extracts are treated with ion-exchange chromatography, adsorption chromatography and gel-filtration chromatography to give high-purity glucokinase. |
