A competitive heterogeneous enzyme immunoassay for detection lactam ring containing antibiotics in milk. The two essential features which permit a more equal competition between the free and conjugate forms of the antibiotic are the use of 1) a purified specific antibody prepared with an immunogen composed of the antibiotic bonded through its lactam ring to a protein and 2) an enzyme antibiotic conjugate formed by the attachment of the enzyme to the antibiotic in a manner which leaves the lactam ring free to compete with antibody. The preparation of the antibody is illustrated in figure 1. The assay scheme is illustrated in figures 4, 7 and 8 The assay is relatively inexpensive and is capable of detecting the antibiotics at a magnitude of 0.01 units per milliliter by relatively untrained personnel. |
