<p>PURPOSE:To prepare a novel enzyme, capable of hydrolyzing casein and inhibiting the activity of glucosyltransferase, having an optimal specific pH value without inhibition by FeSO4, DFP, EDTA, etc., and use the enzyme for prevention and remedy of dental caries. CONSTITUTION:A micoorganism belonging to the genus Streptomyces, e.g. Streptomyces globisporus B-1829 strain, is cultivated, and an alkali protease M3 is collected from the culture by the conventional method. The resultant alkali protease M3 is capable of hydrolyzing casein and inhibiting the activity of glucosyltransferase, has an optimal pH of 9-12.5 in the case of casein as a substrate, and retains 50% or more protease activity after standing at pH 4-9 and 37 deg.C for 6 days. The optimal temperature is 65 deg.C and will not be inhibited by iodoacetic acid, DFP, EDTA and FeSO4 of 10<-3>M, and has a molecular weight of about 1.7X10<4> and an isoelectric point of about pH9.5.</p> |
