1475586 N - Hydroxyamidine derivatives AKZO NV 17 July 1974 [3 Aug 1973] 31745/74 Heading C2C Novel compounds of Formula I or pharmaceutically acceptable acid addition salts thereof, in which B represents a N- hydroxy-amidino group of the general formula A represents a methylene-, ethylene- or propylene group, optionally substituted with C 1-6 alkyl, r is the number 0, 1, 2, 3 or 4, each R 1 individually represents hydroxy, C 1-6 alkyl, C 1-6 alkylthio, C 1-6 alkoxy, halogen (F, Cl, Br, I), trifluoromethyl, nitro, amino or alkylenedioxy (1-4 C), C n H 2n is an alkylene group with 0-4 carbon atoms, R 2 represents hydrogen, C 1-6 alkyl, aryl or aralkyl, R 3 , R 4 , R 5 each individually represent hydrogen, C 1-6 alkyl, aralkyl or an acid residue and R 6 represents hydrogen, derived from a C 1-18 organic carboxylic acid; C 1-6 alkyl, or phenyl- or benzyl optionally substituted with methyl, are prepared by (1) reacting the corresponding nitrile of Formula II with hydroxylamine or a derivative thereof of formula: HNR 6 -OR 3 , or an acid addition salt thereof, (2) condensing an O- or S-alkyliso(thio) amide of Formula IV or an acid addition salt thereof [X=-O- or -S- and R 0 = C 1-6 alkyl] with hydroxylamine or a derivative of the above formula or an acid addition salt thereof, (3) reacting a compound of Formula IVA with said hydroxylamine or derivative thereof [R 6 = H], or (4) reacting a compound of Formula VI [Hal represents F, Cl, Br or I] with NH 3 or a primary or secondary amine of formula, HNR 4 R 5 . Also, the N-hydroxy group of a compound I (with R 3 =H) may be (ar)alkylated or acylated and a nitrogen atom of a compound I, in which at least one of R 4 , R 5 and R 6 is hydrogen, may be acylated or (ar)alkylated. The examples prepare the 1-carboxamidoximes and derivatives thereof of benzocyclobutene, indane and 1,2,3,4-tetrahydronaphthalene. Acid addition salts prepared are those with HCl and maleic acid. Example II describes the optical resolution of racemic-benzocyclobutene-1-carboxamidoxime using L(-)- mandelic acid. The compounds of Formula I and their acid addition salts possess antihypertensive and vasodilatory activities, anti-thrombosis activity and a general biocidal activity. |
