PURPOSE:To obtain the titled enzyme useful for quantitative determination of acetic acid industrially advantageously, by cultivating a heat-resistant acetic acid kinase producing bacterium belonging to Bacillus stearothermophilus in a nutrient medium, followed by separating and collecting the titled enzyme from the culture. CONSTITUTION:Strain UK788(FERM-P 5141) belonging to Baillus stearothermophilus is cultivated aerobically in a nutrient medium at about 20-80 deg.C for about 2-6hr. A mold is collected from the culture by a method, e.g., centrifugation, filtration, etc., pulverized and destroyed, and entrifuged to give a supernatant liquid. The liquid is purified by adding an organic solvent or a salt, followed by fractionation or by adsorbing the liquid on a carrier, to give the desired heat-resistant acetic acid kinase. |
