Heading C2C. 51-Nucleotides are prepared by reacting the corresponding nucleotides with phosphoryl chloride and/or tetrachloropyrophosphate in the presence of a trialkyl or trihaloalkyl phosphate and hydrolysing the nucleoside-51-phosphorochloridate so formed. The nucleoside need not be protected in the 21- and 31-positions. An acid may be present and may be formed in situ by reaction of the phosphorylating agent with added water or an alcohol. Examples prepare sodium adenosine-51 phosphate, xanthosine-51- phosphate, 6 - chloro - 9 - # - D - ribofuranosyl purine - 51 - phosphate, uridine - 51 - phosphate, 2 - methylmercaptoinosine - 51- phosphate, inosine - 51 - phosphate and its Na salt, guanosine - 51 - phosphate and its Na salt, (disodium) desoxyinosine - 51 - phosphate, 5 - amino - 4 - carbamoyl - 1 - # - D - ribofuranosyl - imidazole - 51 - phosphate. Some adenosine - 31(21),51 - diphosphate and -21(31) - phosphate, and desoxyinosine - 31,51 - diphosphate are also obtained in Examples 13 and 14 respectively.
