<p>Isolation of a plasminogen activator comprises (a) selective adsorption of the activator on a support having specific affinity for it and formed of soluble fibrin fragments covalently joined to an insoluble matrix; and (b) elution of the activator from the support. The prefd. insoluble matrix is a dextran polymer activated by cyanogen bromide or a porous glass e.g. aminated porous silica.. The activator is used for treatment of thromboses. This is an improved method compared with that of FR applications 73-042981 (Derwent 27490W/17) and 75-013932 (Derwent 84971X/46). It allows large scale production of the activator using less complex (fewer stages) prodn. methods, and gives a prod. having excellent purity. 20-50 million units are isolated from 100 kg of fresh animal organs; and the product has activity of 15,000-20,000 units per mg of protein.</p> |
