PURPOSE: To obtain the subject preparation in simple operations by subjecting transferrin-containing plasma fractions to inactivation treatment in combination of β-propiolactone treatment with UV light irradiation, and then to ion- exchanging chromatography. CONSTITUTION: This preparation of highly pure and biologically active transferrin protected from virus is to treat transferrin-containing plasma fractions with ammonium sulfate, polyethylene glycol, etc., remove immunoglobulin by precipitation reaction, remove the precipitant from the residual solution by ultrafiltration and gel filtration, adjust the residue to 0.005-0.5 mole/l ionic concentration and 10-60g/l protein concentration, then, treat with 5% β-propiolactone at pH 6-9, dilute the treated solution to 2-25g/l protein concentration, treat the continuous flow with UV rays at 254nm, subject the treated solution to ion-exchanging chromatography in an ionic strength to absorb all proteins except transferrin and finally concentrate, sterilize and filter the solution. |
