PURPOSE:To provide a method for judging the base sequence of a DNA, not requiring the preparation of the gel for the DNA unlike conventional methods, not needing a primer elongation reaction with a DNA polymerase and the cleavage of the DNA with a special restriction enzyme, capable of easily performing an analysis for the judgment, and not necessary for the labeling of the DNA with a radioactive isotope. CONSTITUTION:This method for judging the base sequence of a DNA comprises individually successively cleaving the bases of the double strand DNA composed from single strands having (n) bases from its molecular end to obtain the mixture of double strand nucleotides whose one strands each has from (n-1) bases to one base, respectively, splitting the double strand nucleotides, selecting either of nucleotides on the sense strand side or the antisense strand side, measuring the respective quantities of the mixture of the selected single strand nucleotides, arranging the single strand nucleotides in the order of the quantities, and measuring the differences between their quantities to judge the sequence of the bases of the selected strand, in its turn, the corresponding strand. |
