NEW MATERIAL:A glycerolkinase having following properties: the mode of action: as given in the equation, molecular weight: 72,000+ or -7,200, isoelectric point: 4.5, Km value: glycerol 4.8X10<-5>M, dihydroxyacetone 6.6X10<-4>M, D-glyceraldehyde 3.5X10<-4>M, ATP 2X10<-4>M,specificity to nucleotides: ATP>CTP>ITP>>GTP, UTP, optimum pH: 9-10, pH stability: 5.5-10, influence by metal ions: Mg ion increases and Ca and Mn ions hinder the action, thermal stability: keeping stably physical and chemical properties up to 45 deg.C. USE:Diagnostic enzyme and others. PREPARATION:A bacterium producing glycerolkinase such as Streptomyces canus A2408 is inoculated in a culture containing carbon and nitrogen sources and inorganic salts and cultured at 25-30 deg.C for 1-3 days. Then, the glycerolkinase formed is collected from the culture mixture.
