PURPOSE:To produce cytochrome P-450 by inoculating a strain of Streptomyces carbophilus SAND62585 is a liquid medium which contains appropriate carbon and nitrogen sources and is sterilized in a usual manner and culturing the strain. CONSTITUTION:A strain of Streptomyces carbophilus SANK 62585 is inoculated in a liquid medius containing appropriate amounts of appropriate carbon and nitrogen sources, sterilized and kept at 5-8 pH. 4 hours to 3 days after inoculation, an appropriate inducer is added to the culture mixture in an amount of 1-5 mM to effect the cultivation for 2 hours to a week. The cell bodies obtained are crushed in a buffer solution with ultrasonic waves, centrifuged, and the resultant crude enzyme solution is subjected to adsorption and elution with ion-exchange column chromatography. Thus, electrophoretically homogeneous peaks, P-450aca-1, P-450aca-2, and P-450aca-3 are obtained. |
