<p>Prodn. of supporting ampholytes for use in isoelectric focusing is carried out by (a) reacting an amino cpd. (I) with a bifunctional reagent (II) capable of dimerising or telomerising (I), and (b) introducing acid functions into the produce by reacting with a suitable organic acid or analogous reagent. Specifically, (I) is an amino acid, a monoamine, or a polyamine with a small no. of N atoms, 2 of which are sepd. by a chain of the type (CH2)2, (CH2)3, CH2CH(OH)CH2 or CH2CH(OH)CH(OH)CH2. The ampholytes provide highly linear pH gradients (e.g. 1-3% deviation), thus ensuring high resolution (e.g. 0.001 unit over the whole pH range in polyacrylamide gel).</p> |
