PURPOSE:A protease that is produced by a microorganism belonging to Achromobacter and spefically cleaves the carbonyl side of lysine is employed to synthesize peptides and pepetide derivatives. CONSTITUTION:In the presence of a protease that is produced by Achrombacter lyticus and specifically cleaves the carbonyl side of lysine, the reaction between an amino acid derivative, peptide or peptide derivative of formula I (X is protecting group for alpha amino acids, residue of amino acid or peptide residue; A is L- lysine residue; Y is hydroxyl, amino, alkoxy, etc.) and another amino acid derivative, peptide or peptide derivative of formula II (B is amino acid residue, peptide residue; Z is hydroxyl, amino, alkoxy, etc.) is conducted at a pH ranging from 5- 9 or from 8-11 at a moear ratio of 1/1-1/100 to synthesize a peptide derivative of formula III (X, A, B and Z are the same as the above).
