An assay for substances demonstrating peroxidative activity is based on the peroxidase catalyzed oxidation of dichlorofluorescin to dichlorofluorescein by peroxide. The assay composition comprises diacetyldichlorofluorescin and a source of hydrogen peroxide. The dichlorofluorescin is formed in situ from diacetyldichlorofluorescin. Immunoassays for antigens or antibodies using species labelled with a substance having peroxidative activity may be performed.
