PURPOSE:To prepare a high purity alpha-cyclodextrin in high yield, by first treating a specific cyclodextrin-glycocyltransferase (CG-tase) with a mixture of alpha-cyclodextrin and a straight-chain oligosaccharide having specific composition, under specific pH range, and then reacting with a specific sacchrified alpha-amylase. CONSTITUTION:A CG-tase obtained by culturing alkaliphilous bacteria belonging to Bacillus genus, is reacted with a mixed solution containing 100 parts of alpha-cyclodextrin and 20-80 parts of a straight-chain oligosaccharide such as glucose and maltose, at pH4-11. After heating the reaction liquid to deactivate the CG-tase, it is reacted with a saccharified alpha-amylase obtained by culturing bacteria belonging to Bacillus genus. Since the saccharides existing in the reaction product are only alpha-cyclodextrin and glucose, etc., the cyclodextrin can be easily purified using an organic solvent only once, with little problems on the waste water disposal. |
