PURPOSE:To prepare dopamine almost quantitatively, by treating L-dopa with a specific enzyme which catalyzes the conversion of L-dopa to dopamine. CONSTITUTION:About 0.1-1% of L-dopa is added to a conventional medium containing a carbon source (glucose, etc.), a nitrogen source and minor organic nutrients (peptone, yeast extract, etc.), and an inorganic salt (Mg salt, etc.). Bacteria belonging to Streptococcus and capable of producing L-dopamine from L-dopa, are inoculated in the medium and cultured at about pH5-8 and 25-40 deg.C. The cultured liquid or living cells of the bacteria separated from the liquid, are added to L-dopa to produce L-dopamine which can be separated, e.g. by filtering the reaction liquid to remove the cells and other insoluble substances, and treating the filtrate with an ion exchange resin, etc. |
