A process for producing a substance by culturing microorganisms comprises pre-culturing the microorganisms and then inoculating the resulting pre-culture broth into a main culture initial medium. The total concentration of carbohydrate and hydrocarbon oxidation product in the main culture initial medium is less than 0.4% by weight in terms of the carbon equivalent. Then the microorganism is cultured by adding a carbohydrate and/or a hydrocarbon oxidation product after the microorganisms have begun logarithmic growth. The microorganisms are microorganisms belonging to the genus Mortierella. The substance is a highly unsaturated fatty acid. |
