PURPOSE:To isolate glucose isomerase in high efficiency, by disintegrating a glucose isomerase-producing bacteria belonging to Actinoplanes genus with a high-pressure homogenizer. CONSTITUTION:A glucose isomerase-producing bacteria belonging to Actinoplanes genus, e.g. Actinoplanes missouriensis (NRRL-B3342), etc., is cultured by conventional process, and the resulting slurry containing 0.2-1.2% of bacterial cells is treated with a high-pressure homogenizer under a pressure of 10-800 kg/cm<2> to disintegrate the cells. The fragments of the cell are filtered, and the isolated glucose isomerase is adsorbed and immobilized with a strong basic anion exchange resin at pH5.5-9.5 and 0-70 deg.C. The adsorption rate is extremely high because the glucose isomerase activity of the enzyme-containing liquid is as high as 200 U/ml. |
