摘要 |
PURPOSE:To attain a cell adhesion activity comparable to that of human fibronectin(FN) by compounding a specified amino acid sequence. CONSTITUTION:A peptide composed of 283 amino acid residues and originated from FN is modified and prepared by genetic engineering technique and the Ser<1496> of the RGDS sequence which is the minimum active unit for the cell adhesion of the piptide composed of 279 amino acid residues (Pro<1239>-Met<1517>) is converted to Val to obtain a polypeptide (A) having the amino acid sequence of formula and having cell adhesion activity. A plasmid pTFD707 coding the component A is introduced into E.coli BW313 to obtain a recombinant plasmid pTFD707-V (B). The component B is introduced into E.coli JM109 and the transformant E.coli JM109/pTF7321 (C) is cultured in a medium to obtain a cultured product (D). The microbial cells are collected from the component D and obtained cells are extracted and purified by ultrasonic treatment, dialysis, etc., to obtain the objective polypeptide having cell adhesion activity. |
